README for Figure6B_TPM.xlsx

*** This file contains the raw data obtained on DNA and DNA-HLp complexes using tethered particle motion represented in Figure 6B of

Legend Figure 6B: TPM experiment with HLp and 685 bp DNA. Each measurement point represents the average of triplicate measurements, with error bars indicating standard deviations. The line connecting the points was generated by fitting the means to a logistic function.

*** The data were obtained using Tethered Particle Motion as described in the associated article.

*** Data obtained at each HLp concentration is given on an individual sheet.
Each individual sheet reports on individual tethered DNA molecules or protein-DNA complexes. Each row contains the data of one bead/tether.
Column A: root mean square in nm
Column B: anisotropy = anisotropic ratio (movement in x-direction/movement in y-direction)
Column C: stdev = standard deviation
Column G: Fit parameters = The mean and standard deviation values obtained from fitting a normal distribution to the root mean square values (column A)

The values given in column B and C are tether quality criteria and used for data filtering as described in the associated article.
Histograms showing the distribution of the root mean square values are shown together with the fit and a kernel density estimator.
