In the first video, the multiwell MEA has been used to test the temperature variation during the period of US-ON. A thermocouple type K was placed at the bottom of the well and the ultrasound transducer immersed in the well. The input voltage was the same as the one used during the cell experiment and the MEA plate was placed on top of a hot plate to reproduce the experimental conditions. After 60 seconds of ultrasound the temperature increase was 0.8 °C.
In the second video, the heat profile generated by the ultrasound transducer used for the ultrasound neuromodulation experiment is observed using the Schlieren system. The trasnduecr is placed in the same Schlieren setup configuration used to validate the ultrasound field and a thermocouple is placed close to the tip of the transducer. The ultrasound is turned on for 60 seconds with a driving voltage 4.5 times higher than the one used during the cell experiment. It can be seen that the heat profile generated by the trasducer remains rather shallow the tip of the transducer and the water surface, suggesting that the heat does not reach the neuronal culture and it is dispersed at the liquid surface, rather than towards the neurons.
